About Palmitoyl Tripeptide-5
Palmitoyl tripeptide-5 (syn-Coll; sequence: Pal-Lys-Val-Lys) is a fatty acid-conjugated synthetic tripeptide that stimulates collagen synthesis by mimicking the activity of thrombospondin-1 (TSP-1), an endogenous activator of transforming growth factor-beta (TGF-β). TGF-β is the primary regulatory cytokine for extracellular matrix remodeling — upregulating collagen I, III, and IV production in dermal fibroblasts. The palmitoyl lipid tail dramatically improves skin penetration by increasing lipophilicity and enabling passage through the stratum corneum barrier. In vitro fibroblast studies demonstrate 350% increases in collagen production relative to untreated controls at research concentrations. Clinical evidence includes randomized double-blind trials showing statistically significant reductions in wrinkle depth and skin surface roughness after 84 days of topical application in formulations containing 4–8 ppm of active peptide. Unlike retinoids, palmitoyl tripeptide-5 causes no photosensitivity, irritation, or barrier disruption — making it suitable for sensitive skin and daily use. Unlike matrix metalloprotease (MMP) inhibitors, which prevent collagen breakdown, this peptide works anabolically to stimulate new matrix production.
